中文摘要: 硝酰基或亚硝酰氢(Nitroxyl, HNO),是一氧化氮(NO)单电子还原产物的质子化形式。HNO作为一种活性氮物种,却表现出与NO截然不同的化学反应活性,因其在许多生物功能和病理活性中的独特作用而备受关注。尽管对HNO化学生物学的认识已取得了很大进展,但由于HNO具有反应性高,寿命短的特点,使其检测仍具有很强的挑战性,因此开发可靠的方法来检测HNO至关重要。近年来,荧光探针因其灵敏度高、操作简单、实时监测、无创等优点被广泛用于生命科学等领域,用于检测HNO的荧光探针也取得了非常大的进展。本文总结了目前已有的四种HNO荧光探针的设计类型,重点介绍了以二苯基膦基为识别位点的荧光探针的研究进展,为日后设计新型HNO探针提供参考,并对该领域所面临的挑战与发展进行了展望。
Abstract:Nitroyl or nitrosoyl hydrogen (Nitroxyl, HNO) is the protonated form of the single electron reduction product of nitric oxide (NO). As a reactive nitrogen species, HNO exhibits a distinct chemical reactivity from NO, attracting much attention for its unique roles in many biological functions and pathological activities. Although great progress has been made in understanding the chemical biology of HNO, HNO is still highly challenging due to its high reactivity and short life span, so it is crucial to develop reliable methods to detect HNO. Recently, fluorescent probes have been widely used in life sciences and other fields due to their high sensitivity, simple operation, real-time monitoring and non-invasive advantages, and those for detecting HNO have also made great progress. This paper summarized four existing design types of HNO fluorescent probes, focusing on the progress of fluorescent probes with the diphenylphosphonic group as the recognition site, to provide a reference for the future design of new HNO probes, the challenges and developments in the field are also prospected.
