中文摘要: 为了解甘肃礼县所产大黄中无机元素含量情况,测定大黄中铅(Pb)、砷(As)、镉(Cd)、铬(Cr)、汞(Hg)、镍(Ni)、铜(Cu)、锌(Zn)、硒(Se)、钾(K)、钙(Ca)、钠(Na)、镁(Mg)、磷(P)和铁(Fe)等15种无机元素含量,并分析元素含量与种植年限的相关性,及对大黄中重金属及有害元素进行健康风险评估。采用微波消解前处理,采用电感耦合等离子体串联质谱法(ICP-MS/MS)和电感耦合等离子体发射光谱(ICP-OES)测定大黄样品中无机元素的含量,并进行相关性分析和偏最小二乘判别(OPLS-DA)分析,根据测定结果计算重金属及有害元素的最大耐受量(EDI)、靶标危害系数(THQ)和致癌风险(CR)进行健康评估。88批次大黄样本中Pb、As、Cd、Hg、Cu均未超出2020版《中国药典》规定的限值,相关性分析表明,Pb、As、Cd、Cr、Ni、Cu、Zn、Na、Mg、Fe含量与年份存在显著的相关性;最小二乘判别法分析可以较好的区分礼县不同种植年限的大黄无机元素含量;健康风险评估结果表明大黄样本总体上是安全的。礼县所产大黄未对使用人群造成健康危害,不存在潜在健康风险。

Abstract:In order to understand the content of inorganic elements in rhubarb produced in Lixian County, Gansu Province, and to determine the contents of 15 inorganic elements in rhubarb, including lead (Pb), arsenic (As), cadmium (Cd), chromium (Cr), mercury (Hg), nickel (Ni), copper (Cu), zinc (Zn), selenium (Se), potassium (K), calcium (Ca), sodium (Na), magnesium (Mg), phosphorus (P), and iron (Fe) and analyze the correlations between elemental contents and years of planting, and to assess the health risks of heavy metals and harmful elements in rhubarb. Microwave digestion was used for the pretreatment, and inductively coupled plasma tandem mass spectrometry (ICP-MS/MS) and inductively coupled plasma emission spectrometry (ICP-OES) were used to determine the contents of inorganic elements in rhubarb samples, and correlation and partial least squares discrimination (OPLS-DA) analyses were carried out to calculate the maximum tolerances (EDI), target hazard coefficients (THC), and maximum tolerances (THC) for heavy metals and harmful elements in rhubarb samples. ), target hazard factor (THQ) and carcinogenic risk (CR) were calculated from the determination results for health assessment. Pb, As, Cd, Hg and Cu in 88 batches of rhubarb samples did not exceed the limit values specified in the 2020 edition of the Chinese Pharmacopoeia. Correlation analysis showed that there was a significant correlation between the contents of Pb, As, Cd, Cr, Ni, Cu, Zn, Na, Mg and Fe and the year; the least squares discriminant analysis could better differentiate between rhubarb inorganic element contents of different planting years in Li County. The results of health risk assessment showed that the rhubarb samples were generally safe.The rhubarb produced in Lixian County does not pose health hazards to the user population, and there is no potential health risk.
