中文摘要: 金属元素的脑区分布对于理解大脑的生理功能、疾病的发生机制以及开发新的治疗策略都具有重要的意义。本研究利用电感耦合等离子体质谱(ICP-MS)和激光剥蚀-电感耦合等离子体质谱(LA-ICP-MS)对大鼠脑中的金属元素进行定量和原位成像分析,元素定量分析结果进行脑区分布特征研究、统计分析和相关性分析。结果表明,大鼠脑中金属元素的含量和分布存在显著差异,这可能与不同脑区的生理功能和代谢活动有关,其中镁、锌元素是脑中含量较丰富的元素,主要分布在胼胝体、尾壳核、侧隔核、纤维边缘以及侧嗅束部分,而铜主要分布在侧隔核、纤维边缘、第三脑室区域。痕量金属铬和镍主要分布在胼胝体。LA-ICP-MS可以对体内元素的空间分布进行原位成像,可以用于研究金属元素在神经系统疾病中的作用,揭示潜在的健康风险。
Abstract:The distribution of metal elements within brain regions closely associated with the physiological functions of the brain, the pathogenesis of diseases, as well as the development of novel strategies for neurological disorders. The present study conducted quantitative analysis and in situ imaging of metal elements in rat brains by using inductively coupled plasma mass spectrometry (ICP-MS) and laser ablation-inductively coupled plasma mass spectrometry (LA-ICP-MS). The results of element quantitative analysis were used to study the brain distribution characteristics, statistical analysis, and correlation analysis. The results demonstrated that there were substantial disparities in both the content and distribution of metal elements in rat brains, and this might be associated with the physiological functions and metabolic activities specific to different brain regions. Magnesium and zinc show higher abundance in the brain, and are predominantly distributed in the corpus callosum, caudoputamen, lateral septal nucleus, fimbria, and lateral olfactory tract. Copper is primarily distributed in lateral septal nucleus, fimbria, and the third ventricle. Trace metals such as chromium and nickel are mainly distributed in corpus callosum. LA-ICP-MS provide in situ imaging to monitor the spatial distribution of elements within the body, which can be harnessed to investigate the role of metal elements in neurological diseases and disclose potential health hazards.
