中文摘要: 为了解巴西坚果中硒形态特征,本研究构建了一种高效液相色谱-电感耦合等离子体质谱(High Performance Liquid Chromatography-Inductively Coupled Plasma-Mass Spectrometry,HPLC-ICP-MS)联用技术的分析方法,用于巴西坚果中硒形态的分析测定。坚果样品经40mg蛋白酶 XIV 和12mLTris-HCl 缓冲溶液处理,超声提取3h后,采用 C18 反相色谱柱,以10mmol / L 柠檬酸与 5mmol / L 己烷磺酸钠(pH = 4. 0,含 1%甲醇)作为流动相进行等度洗脱后采用ICP-MS 测定,在6min内可实现硒形态的有效分离和定量测定。实验结果表明不同硒形态的线性相关系数( r) 均大于 0. 999,方法检出限为0.5 ug/kg~1.3μg / kg。不同浓度水平4种硒形态的加标回收率在 93.5% ~ 117.0%范围内,相对标准偏差(Relative Standard Deviation,RSD%)均低于5%;SeCys2加标回收率在50%左右,推测 SeCys2 稳定性可能与巴西坚果的高蛋白基质等相关。分析欧盟标准物质小麦粉(ERM? BC210a)中SeMet ,测定结果在标识值范围内;研究结果表明建立的分析方法灵敏、快速、准确,适用于巴西坚果中硒形态的分析测定。同时研究发现来自巴西、秘鲁、玻利维亚等地区的巴西坚果中主要硒形态为SeMet和少量SeCys2,其中SeMet含量占硒形态之和的95%。本研究将为富硒坚果类食品营养价值的进一步研究提供参考。
Abstract:In this study, a high performance liquid chromatography-inductively coupled plasma-mass spectrometry (HPLC-ICP-MS) method was established to determine selenium speciation in Brazil nuts. The sample was treated with 40mg protease XIV and 12mLTris-HCl buffer solution, extracted by ultrasonic for 3 hours, and then eluted by C18 reversed-phase chromatographic column with 10mmol/L citric acid and 5mmol/L sodium hexanesulfonate (pH = 4. 0, containing 1% methanol) as mobile phases. The effective separation and quantification of selenium forms could be achieved within 6min. The method verification shows that the linear correlation coefficients (r) of the five selenium speciation methods were all greater than 0. 999, and the detection limits of Se (VI), Se (IV), MeSeCys and SeMet were 0.5 ~ 1.3 μg/kg. The recoveries of four selenium speciation at different concentration levels were 93.5% ~ 117.0%, the relative standard deviation (RSD) was less than 5%, and the recovery rate of SeCys2 was about 50%. It was speculated that the stability of SeCys2 might be related to the high protein matrix of Brazil nuts. The accuracy of the method was verified by analyzing the SeMet in the European Union standard material wheat flour (ERM? BC210a). The method established in this study is sensitive, rapid and reliable, and is suitable for the analysis and determination of selenium forms in Brazil nuts. It is found that the main selenium forms of Brazil nuts from Brazil, Peru, Bolivia and other places are SeMet and a small amount of SeCys2, of which SeMet content accounted for 95% of the sum of selenium forms. It provides reference for further study on nutritional value of selenium-enriched nuts.
