中文摘要: 随着单细胞技术的快速发展,单细胞电感耦合等离子体质谱(SC-ICP-MS)以其高灵敏度与高准确度的优势,已成为表征细胞内元素含量及揭示细胞间元素分布异质性的关键分析技术,有力推动了细胞微量元素研究与细胞异质性解析的深入。本文系统综述了SC-ICP-MS技术的最新研究进展,重点从单细胞进样方法、定量校正策略以及质量分析器优化等方面,探讨其如何提升单细胞分析的通量与数据可靠性,并总结了该技术在生物医学、环境毒理及纳米颗粒细胞摄取等领域的典型应用,以期为相关研究提供方法与参考。

Abstract:With the rapid advancement of single-cell technologies, Single Cell Inductively Coupled Plasma Mass Spectrometry (SC-ICP-MS) has emerged as a key analytical method for quantifying intracellular elemental contents and elucidating cell-to-cell heterogeneity in elemental distribution, owing to its high sensitivity and accuracy. This technique has substantially advanced research on cellular trace elements and the characterization of cellular heterogeneity. This review provides a systematic overview of recent developments in SC-ICP-MS, with a particular focus on innovations in single-cell introduction methods, quantitative calibration strategies, and mass spectrometer optimization. It further summarizes representative applications of SC-ICP-MS in biomedicine, environmental toxicology, and studies of nanoparticle uptake at the single-cell level. Together, these insights aim to offer methodological guidance and perspectives for future developments in this rapidly evolving field.
